絵画や歴史的遺物の表面を解析する際には、最初の状態チェックに実体顕微鏡やズーム顕微鏡が役立ちます。どちらの顕微鏡も長い作動距離と優れた被写界深度を有し、微細構造を低倍率の3Dイメージで観察することができます。
自在なズームポジションによって、織物の個々の繊維を同定したり、絵画の筆のタッチ、亀裂、層、署名を詳細に観察したりすることが可能です。

光学顕微鏡の高度で多彩なコントラスト法によって、最先端の技術を用いた作業が実現します。高感度で非破壊、ほぼ非接触の観察を通して、非常に正確な構造解析と鑑定書の作成に必要なデータ収集が可能となります。
考古学や科学調査のアプリケーションで、織物、絵画、布張り家具などの博物館にある作品の詳細な解析を行う場合には、繊維の違いを区別する必要があります。繊維は多くの場合経年劣化し、断片化、風化、炭化、化石化がみられることもあります。一般的な鑑定や、損傷の評価と原因の解明、修復方法の決定には、慎重な非接触観察が欠かせません。
偏光顕微鏡を使ったHerzog法により天然繊維と合成繊維を区別することで、作品の起源、年代、制作方法について、科学的根拠に基づいた判断が可能となります。顔料の解析からは、絵画に使われている色や修復に用いるべき絵具に関する情報が得られます。また、どの絵具層がオリジナルで、どの絵具層が以前の修復師によって追加されたのかといった詳細も明らかになります。
岩石や鉱物の性質を判断するのには、偏光顕微鏡も最適です。薄片スライドを観察し、岩石の組成や形成の経緯、地質学的起源を知ることができます。偏光顕微鏡をMichel-Lévyの複屈折表と組み合わせることで、肉眼では同定できない有色、無色、あるいは透明の物質について、その薄片の厚み、レターデーション値、干渉色から複屈折量を推定することができます。

X線顕微鏡
マイクロX線CT顕微鏡は、地質学や古生物学の研究、分類、コレクションの一般的な解析に有用で、試料の内部を3Dで非破壊的に観察できます。試料を回転させて複数の方向から撮影することで、外部と内部の構造が3Dモデルとして再構築されます。それを画面上で操作・測定し、実際の大きさ、密度、空孔率、その他のパラメーターを求めます。
修復師は3Dモデルから内部の構造を把握し、肉眼では同等に見える物質を、減衰値の違いによって区別することができます。動物標本などの繊細な試料の場合も、試料調製や切片化によってダメージを与えることなく、内部の構造をそのままの状態でイメージングすることが可能となります。

電子顕微鏡
走査型電子顕微鏡(SEM)では、電子線を使って標本をイメージングします。これにより、マイクロメーター、時にはナノメーターレベルにおける構造の詳細な観察・解析が可能になります。電子顕微鏡は光学顕微鏡よりも高倍率で優れた分解能を備えているため、小さな試料の微細構造を観察することができます。
また、エネルギー分散型X線分析(EDX)検出器と併用することで、絵画の元素組成の同定が可能です。強力な電界放出型SEMでは、コーティングした試料を2 nmの高分解能で観察できます。さらに環境制御型SEMでは、水分除去やスパッタコーティングなしで試料を観察できるため、博物館でのアプリケーションに特に有用です。
