预防、诊断和治疗传染病
微生物学是医学的一个分支,涉及预防、诊断和治疗由细菌、真菌、酵母、寄生虫或病毒等病原体引起的传染病。微生物学家研究病原体的特征、传播方式、感染和生长机制,以设计出治疗方法。
现代细菌学先驱、德国医生兼微生物学家罗伯特·科赫率先通过分离纯细菌培养物鉴定出结核病、霍乱和炭疽病的特定病原体,并因此获得了诺贝尔医学奖。
微生物培养是在实验室中分离传染病的主要方法。测试组织或液体样品是否存在特定病原体,需要通过在选择性或鉴别培养基中的体外生长来确定。培养基可以是固体培养物(大多数为细菌和真菌)、液体培养物(某些寄生虫)或细胞培养物(主要用于病毒检测)。分离出病原体后,可以用光学显微镜对其进行研究。此时,可以通过革兰氏染色等各种非特异性或特异性染色来提高衬度。由于细菌细胞壁的成分不同,通过革兰氏染色还可以鉴别革兰氏阳性和革兰氏阴性细菌。

对显微镜的要求
由于大多数病原体非常小(细菌的平均直径在0.2至2.0微米之间),因此光学显微镜不仅要具有高放大倍率,还需要配备油浸物镜,以便在鉴别球菌(球状)、杆菌(棒状)和螺旋菌(螺旋状)时改善形态学检查。使用荧光显微技术可以在特异性上实现更高灵活性,即可以对结核病进行非特异性鉴定,也可以使用抗体偶联免疫荧光和FISH(荧光原位杂交)进行高度特异性鉴定。
良好区分细菌细胞类型和确保细胞细节清晰可见是进行微生物学研究的绝对先决条件。微生物学家借助高清图像来检查粪便样品中的包囊、原生动物和蠕虫卵、血液样品中的疟原虫或分枝杆菌和其他病原微生物。显微镜检查通常使用明场、相差、暗场、荧光、免疫荧光或FISH显微技术。显微镜的光学质量、随附用于数字记录的相机的保真度以及设备的人体工程学设计可以在筛查患者样品时发挥重要作用。
