评估卵母细胞的质量
在执行胞浆内单精子注射或体外受精程序前,必须评估卵母细胞的质量,为接下来的受精过程进行筛选和准备。
卵母细胞的质量可通过以下体视显微镜检查得到最佳评估:
- 卵母细胞细胞质异常,如空泡化和颗粒化
- 透明带
- 形状异常
- 卵母细胞周围的卵泡细胞呈“日光放射状”
在受精前,必须去除卵母细胞周围的外部体细胞层(透明带除外),同时在体视显微镜的控制下在培养皿中完成剥脱。将卵母细胞反复吸入用于剥脱的移液管中,直至所有外部细胞层脱落。随后检查卵母细胞是否存在形态异常。在卵母细胞采集、剥脱和注射的整个过程中,必须确保约35 - 37.5 °C的恒定温度。因此,解剖体视显微镜可以配备加热台并直接安装在层流箱内。
