Transmission

This impressive cross-section of a microscope objective emphasizes how ZEISS’ sophisticated optics, mechanics and motorization interact in such high-end objective. This high-end microscopy product features a very high numerical aperture, a large working distance, and a water immersion objective imaging living cells (ideal imaging of aqueous samples, correction of spherical errors when imaging from different focal planes by motorized movement of lenses).

The microscope objective transmits the light to a diffusion plate from where it is guided via a fiber into to the CMOS spectrometer CYGUS® (see next step: Detection).
 

Illumination

Colibri 7 from ZEISS is a fast and flexible LED light source that uses up to six different LEDs to generate up to seven fluorescence excitation lines. It is typically used as a light source for gentle fluorescence imaging (fluorescence microscopy), but can also be used for other illumination tasks, e.g. for experiments with specific fluorescent markers that are excited at exactly the right wavelength and require stable and robust illumination for reproducible data and ultra-fast live cell imaging.
The Colibri 7 illumination module emits different wavelengths to a transmissive, beam-focusing element (see next step: Transmission).

Detection

CYGUS® from ZEISS is a high-performance, compact and fiber coupled spectrometer system covering the spectral range from 190 – 1100 nm. It offers great versatility in CMOS detector options with configurations for superior resolution, high sensitivity, and fast readout. It is therefore ideally suited for low-light applications such as fluorescence, emission, absorbance, and reflectance measurements – whether in the laboratory or as an OEM industrial product.
CYGUS® measures the spectrum of the light source Colibri 7 (see first step: Illumination), while it identifies and evaluates wavelengths and relative intensity and visualizes the wavelength behavior in the measurement curve.
